Factor V Leiden is a blood clotting disorder. Factor V is a protein that is needed for blood to clot properly. Factor V Leiden is caused by a change (mutation) in the gene for this protein. The Factor V Leiden protein is harder to regulate than the normal Factor V protein. This makes blood clots more likely to form, a condition called thrombophilia.
The TRUPCR® Factor V Leiden Mutation Testing Kit amplifies and detects the target sequence using allele-specific fluorophore-labelled probes. The target sequence is a single nucleotide guanine/adenine polymorphism in site 1691 (G1691A). Presence of wild-type allele (G1691G) is detected in the HEX fluorescent channel and mutant allele (A1691A) in the FAM fluorescent channel. In case of heterozygous genotype (G1691A) a signal is detected in both channels. The Factor V mutation testing kit contains ready to use master mix and takes advantage of the “hot start” technology, minimising non-specific reactions and assuring maximum sensitivity.
